The compound first appeared in the scientific literature as an investigational agent for type 2 diabetes. Clinical development proceeded through phase 1, phase 2, and phase 3 programs that measured glycemic control as a primary endpoint while recording body weight as a secondary outcome. Regulatory approval in the United States followed in 2022 for glycemic control, and a separate indication for chronic weight management was added later. Subsequent trials have examined cardiovascular outcomes in adults with elevated cardiovascular risk. Debates continue over how much of the observed effect derives from each receptor arm.
Structural work on the molecule centers on a C20 fatty diacid moiety attached through a linker to the peptide backbone. This side chain promotes reversible binding to serum albumin, which slows renal clearance and supports a prolonged action profile. The peptide backbone incorporates aminoisobutyric acid substitutions that limit recognition by digestive enzymes. Together these modifications produce a molecule that is stable enough for subcutaneous delivery but still dependent on careful manufacturing control. Analytical characterization of the active pharmaceutical ingredient typically follows the conventions used for other synthetic peptides.
Tirzepatide is a synthetic peptide composed of 39 amino acids. It acts as a dual agonist at two incretin receptors, the glucose-dependent insulinotropic polypeptide receptor and the glucagon-like peptide-1 receptor. The molecule was designed by modifying the native sequence of glucose-dependent insulinotropic polypeptide to improve metabolic stability and extend its circulation time. Its structure includes several non-natural amino acid residues and a fatty acid side chain. These features distinguish it from earlier single-receptor incretin analogs studied in the same period.
The peptide backbone contains 39 amino acids and includes alpha-aminoisobutyric acid residues, which are not among the standard proteinogenic set. A C20 fatty diacid moiety is attached through a linker, allowing the compound to bind serum albumin and extend its circulation time. This albumin binding is the main reason the molecule supports once-weekly administration rather than more frequent dosing. The measured molecular mass is approximately 4,813 daltons, placing it firmly in the peptide rather than small-molecule class.
Tirzepatide is a synthetic peptide that activates both the glucose-dependent insulinotropic polypeptide (GIP) and glucagon-like peptide-1 (GLP-1) receptors. This dual agonist profile distinguishes it from earlier incretin-based compounds that act on a single receptor. The molecule was engineered from the native GIP sequence and carries several non-natural residues that slow enzymatic breakdown. Researchers designed it to combine the insulinotropic effects of GIP signaling with the appetite and gastric-emptying effects associated with GLP-1 activation.
Development of tirzepatide took place under a research program that sought to test whether simultaneous engagement of two incretin receptors would produce greater metabolic effects than single-receptor agonism. Clinical trials were organized into the SURPASS series for type 2 diabetes and the SURMOUNT series for obesity and weight management. Regulatory clearance for type 2 diabetes came in 2022 in the United States, followed by approval for chronic weight management in 2023. The trial programs reported reductions in glycated hemoglobin and body weight relative to comparators, though long-term cardiovascular and durability data continue to accumulate.

Degradation pathways for tirzepatide include deamidation, oxidation, and aggregation, which are common for therapeutic peptides. These processes can be monitored by size-exclusion chromatography (SEC) for aggregates and ion-exchange chromatography for charge variants. Forced degradation studies under acidic, basic, oxidative, and thermal stress help identify potential impurities. The exact stability profile depends on formulation, concentration, and container-closure system.
Analytical characterization of tirzepatide typically employs reversed-phase high-performance liquid chromatography (RP-HPLC) for purity assessment and peptide mapping. Mass spectrometry, often coupled with electrospray ionization, confirms molecular weight and sequence integrity. Amino acid analysis and capillary electrophoresis may also be used to detect impurities or degradation products. These methods are essential for batch release and stability studies.
Lyophilized material is generally held at -20 degrees Celsius or lower, desiccated and protected from light, where it remains stable for extended periods. Reconstituted or ready-to-use solution is usually kept at 2 to 8 degrees Celsius with minimal agitation. Repeated freeze-thaw cycles should be avoided because they promote aggregation and reduce the soluble monomer fraction. Shipment of frozen solid commonly uses dry ice, while refrigerated liquid moves with validated cold packs. Stability beyond documented periods is not established.
Characterization of the peptide relies on reversed-phase high-performance liquid chromatography for purity and related-substance profiling, with ultraviolet detection near 214 nanometers. Mass spectrometry confirms molecular mass and reveals modifications such as oxidation or deamidation. Peptide mapping after enzymatic digestion verifies the amino acid sequence, while amino acid analysis supplies compositional data. Circular dichroism and infrared spectroscopy are used to assess secondary structure, particularly the alpha-helical content that influences aggregation behavior in solution.
Common degradation routes include hydrolysis of labile amide bonds, deamidation of asparagine and glutamine residues, oxidation of methionine and tryptophan, and non-covalent aggregation. Aggregates can form during freeze-thaw cycling, at elevated pH, or when peptide concentration is high. Each route produces characteristic chromatographic or mass shifts that are tracked during stability studies. Whether a given minor impurity alters biological activity is often an open question, and specification limits are typically set on identity and purity rather than on functional data for trace species.
=== Prescription trends === Between 1998 and 2017, along with imipramine, amitriptyline was the most commonly prescribed first antidepressant for children aged 5–11 years in England. It was also the most prescribed antidepressant (along with fluoxetine) for 12- to 17-year-olds.
Hamas is a prescribed terrorist group in the UK, Israel and many other countries. The film is subsequently removed from BBC iPlayer. Nigel Farage gives up ownership of Reform UK, the party he founded as a private limited company in 2018, and relinquishes his shares in the organisation. Nathan Gill, the former leader of Reform UK in Wales, is charged with offences relating to bribery and is scheduled to appear in court on Monday 24 February. 21 February – The Parliamentary Commissioner for Standards launches an investigation into former minister Andrew Gwynne over the alleged sending of offensive messages through a WhatsApp group. Scottish Labour leader Anas Sarwar says he will deliver the "biggest reform of the NHS in decades" if his party wins the 2026 Scottish Parliament election. Wales's Social Justice Secretary, Jane Hutt, who is the UK's longest serving minister after service in successive Welsh Governments since 1999, announces she will retire from the Senedd in 2026. Sinn Féin confirms it will boycott St Patrick's Day events at the White House over US President Donald Trump's stance on the Israel-Gaza conflict. 24 February – Suspended Labour MP Mike Amesbury is sentenced to ten weeks in prison after admitting to punching a man to the ground in his Cheshire constituency. Westminster's Strangers' Bar is scheduled to reopen following a safety review. The establishment will have CCTV and extra security guards, while bar staff will be trained to look out for any potential safety issues.
The lung's alveoli are not tiny balloons that expand and contract under positive pressure following the Young–Laplace equation, as is taught in some physiology and medical textbooks. The tissue structure is more like a sponge with polygonal spaces that unfold and fold under negative pressure from the chest wall. Half of body heat is not lost through the head, and covering the head is no more effective at preventing heat loss than covering any other portion of the body. Heat is lost from the body in proportion to the amount of exposed skin. The head accounts for around 7–9% of the body's surface, and studies have shown that having one's head submerged in cold water causes a person to lose only 10% more heat overall. Adrenochrome is not harvested from living people and has no use as a recreational drug. Hunter S. Thompson conceived a fictional drug of the same name in his book Fear and Loathing in Las Vegas, apparently unaware that a real substance by that name existed; it is Thompson's fictional adrenochrome, and not the real chemical compound, that is the source of numerous conspiracy theories revolving around human trafficking to harvest the fictional drug. Men and women have the same number of ribs: 24, or 12 pairs. The erroneous idea that women have one more rib than men may stem from the biblical creation story of Adam and Eve. The use of cotton swabs (a.k.a. cotton buds or Q-Tips) in the ear canal has no associated medical benefits and poses definite medical risks.
== Society and culture == In some parts of Africa, including Somalia, Ethiopia and Eritrea, the uvula or a section of it is ritually removed by a traditional healer. In this case, the uvula may be noticeably shortened. It is not thought to contribute to velopharyngeal inadequacy, except in cases where the tonsils have also been removed.
This constriction was found to be competitively antagonized by prazosin (a standard α1 antagonist) and ketanserin, with prazosin being the more potent antagonist (pA2 = 9.38, vs pA2 = 8.23 for ketanserin). Synephrine constrictions were also antagonized by BRL-15,572, but not by SB-216,641 (used here as a selective 5-HT1B antagonist), or by propranolol (a common β antagonist). In studies on guinea pig atria and trachea, Jordan and co-workers also found that synephrine had negligible activity on β1 and β2 receptors, being about 40000x less potent than norepinephrine. Experiments with cultured white fat cells from several animal species, including human, by Carpéné and co-workers showed that racemic synephrine produced lipolytic effects, but only at high concentrations (0.1-1 mM). The potency, expressed in terms of pD2 of synephrine in these species was as follows: rat: 4.38; hamster: 5.32; guinea pig: 4.31; human: 4.94. In comparison, isoprenaline had a pD2 = 8.29 and norepinephrine had pD2 = 6.80 in human white fat cells. The lipolytic effect of 1 mM/L of synephrine on rat white fat cells was antagonized by various β-antagonists with the following inhibitory concentrations (IC50): bupranolol: 0.11 μM; CGP-20,712A (β1 antagonist): 6.09 μM; ICI-118,551 (β2 antagonist): 3.58 μM; SR-5923A (β3 antagonist): 17 μM. The binding of racemic synephrine to cloned human adrenergic receptors has been examined: Ma and co-workers found that synephrine bound to α1A, α2A and α2C with low affinity (pKi = 4.11 for α1A; 4.44 for α2A; 4.61 for α2C).

Modern definitions are concerned with the fundamental chemical reactions common to all acids. Most acids encountered in everyday life are aqueous solutions, or can be dissolved in water, so the Arrhenius and Brønsted–Lowry definitions are the most relevant. The Brønsted–Lowry definition is the most widely used definition; unless otherwise specified, acid–base reactions are assumed to involve the transfer of a proton (H+) from an acid to a base. Hydronium ions are acids according to all three definitions. Although alcohols and amines can be Brønsted–Lowry acids, they can also function as Lewis bases due to the lone pairs of electrons on their oxygen and nitrogen atoms.
On April 5, the Iraqi government announced "the complete crushing of acts of sedition, sabotage and rioting in all towns of Iraq." An estimated 25,000 to 100,000 Iraqis were killed in the uprisings. In Kuwait, the Emir was restored, and suspected Iraqi collaborators were repressed. Eventually, over 400,000 people were expelled from the country, including a large number of Palestinians, because of PLO support of Saddam. Yasser Arafat did not apologize for his support of Iraq, but after his death Mahmoud Abbas formally apologized in 2004 on behalf of the PLO. This came after the Kuwaiti government formally forgave the group. There was some criticism of the Bush administration, as they chose to allow Saddam to remain in power instead of pushing on to capture Baghdad and overthrowing his government. In their co-written 1998 book, A World Transformed, Bush and Brent Scowcroft argued that such a course would have fractured the alliance, and would have had many unnecessary political and human costs associated with it. In 1992, the US defense secretary during the war, Dick Cheney, made the same point:

=== Immunity === RNAi is a vital part of the immune response to viruses and other foreign genetic material, especially in plants where it may also prevent the self-propagation of transposons. Plants such as Arabidopsis thaliana express multiple Dicer homologs that are specialized to react differently when the plant is exposed to different viruses. Even before the RNAi pathway was fully understood, it was known that induced gene silencing in plants could spread throughout the plant in a systemic effect and could be transferred from stock to scion plants via grafting. This phenomenon has since been recognized as a feature of the plant immune system which allows the entire plant to respond to a virus after an initial localized encounter. In response, many plant viruses have evolved elaborate mechanisms to suppress the RNAi response. These include viral proteins that bind short double-stranded RNA fragments with single-stranded overhang ends, such as those produced by Dicer. Some plant genomes also express endogenous siRNAs in response to infection by specific types of bacteria. These effects may be part of a generalized response to pathogens that downregulates any metabolic process in the host that aids the infection process. Although animals generally express fewer variants of the Dicer enzyme than plants, RNAi in some animals produces an antiviral response. In both juvenile and adult Drosophila, RNAi is important in antiviral innate immunity and is active against pathogens such as Drosophila X virus. A similar role in immunity may operate in C.

=== Catabolism === Catabolism of cardiolipin may happen by the catalysis of phospholipase A2 (PLA) to remove fatty acyl groups. Phospholipase D (PLD) in the mitochondrion hydrolyses cardiolipin to phosphatidic acid.
=== Initial stranding === The carcass was first spotted on the evening of November 30, 1896, by two young boys, Herbert Coles and Dunham Coretter, while bicycling along Anastasia Island. The enormous mass was half buried in the sand, having sunk under its immense weight. The two boys thought the carcass was the remains of a beached whale, as a similar stranding had occurred two years earlier near the mouth of the Matanzas River, located several miles to the south of St. Augustine (see map). The two boys returned to St. Augustine the same day and reported their discovery to a local physician, Dr. DeWitt Webb. Webb, who was the founder of the St. Augustine Historical Society and Institute of Science, came to the beach the following day, December 1, to examine the remains. He would be the only known person of an academic background to see the specimen in situ. His first impression was that it was the remains of an animal, very mutilated, and in an advanced state of decomposition. The carcass was very pale pink, almost white, in colour, with a silver reflection in the sunlight. It was composed of a rubbery substance of a very hard consistency, such that it could only be cut with great difficulty. The part of the carcass that was visible measured 18 feet (approx. 6 meters) in length and 7 feet (approx. 2+1⁄2 meters) in width. Webb estimated its weight at nearly 5 tons, if not more. He believed it was the remains of a giant octopus, as it appeared to have the stumps of four arms, with another arm buried nearby.
=== Incidents === The 1981 Louisville sewer explosions, which destroyed over 13 mi (21 km) of sewer lines and streets in the Kentucky city, were caused by ignition of hexane vapors which had been illegally discharged from a soybean processing plant owned by Ralston-Purina. Hexane was attributed as the cause of an explosion that occurred in the National University of Río Cuarto, Argentina on 5 December 2007, due to a hexane spill near a heat-producing machine that exploded, producing a fire that killed one student and injured 24 more. Occupational hexane poisoning has occurred with Japanese sandal workers, Italian shoe workers, Taiwan press proofing workers, and others. Analysis of Taiwanese workers has shown occupational exposure to substances including n-hexane. In 2010–2011, Chinese workers manufacturing iPhones were reported to have suffered hexane poisoning.
== As food == Partly in consequence of its size, the giant gourami is a significant food fish, and in its native regions it has been harvested as a customary food source. In Southeast Asian cuisine, gourami is highly valued as food due to its thick flesh, texture and flavour. Unlike carp and milkfish, gouramis do not have fine bones within their flesh, which has increased their market value. In Southeast Asian market, gourami is one of the most highly-valued freshwater food fish. Gourami flesh is rich in protein and minerals. It is a popular food fish in Indonesian, Malaysian and Thai cuisines. Gouramis are particularly popular in Sundanese cuisine of Indonesia, where they often being fried as ikan goreng, grilled as ikan bakar or cooked with spice inside a banana leaf wrap as pepes. In some parts of Southeast Asia, they are salted to preserve and prolong shelf life.

It activates both the glucose-dependent insulinotropic polypeptide receptor and the glucagon-like peptide-1 receptor. This dual activity separates it from agents that act on only one of the two receptors. The relative contribution of each receptor to clinical effects remains an open area of study.
United States approval for type 2 diabetes was granted in 2022, and a chronic weight management indication was added later. Approval timelines differ across other jurisdictions. Earlier human data came from phase 1 and phase 2 trials published before those decisions.
Native hormones are short-lived peptides that enzymes degrade within minutes. Tirzepatide carries non-natural residues and a fatty acid chain that resist this breakdown and extend circulation time. The result is a longer interval between administrations than the natural hormones could support.
It binds and activates both the GIP and GLP-1 receptors, making it a dual incretin receptor agonist. Single-receptor GLP-1 agonists act on one target only. The dual profile is the defining pharmacological feature of the molecule.
