Tirzepatide is a synthetic linear peptide of 39 amino acids that acts as a dual agonist at the glucose-dependent insulinotropic polypeptide (GIP) and glucagon-like peptide-1 (GLP-1) receptors. Its sequence derives from native GIP but incorporates non-natural residues and a C20 fatty diacid moiety linked to a lysine side chain. The lipophilic chain promotes albumin binding, which slows renal clearance and extends circulation time. The unmodified peptide has a molecular formula of C225H348N48O68 and a molecular mass near 4,813 daltons.
Receptor activation by tirzepatide raises intracellular cyclic AMP through Gs-coupled signalling at both targets. At the GLP-1 receptor the downstream effect includes glucose-dependent insulin release, suppressed glucagon secretion, delayed gastric emptying, and reduced appetite signalling in the hypothalamus. GIP receptor engagement adds insulinotropic activity and appears to influence lipid handling in adipose tissue. Because both receptors are stimulated at the same time, the pharmacological profile differs from that of selective GLP-1 receptor agonists, and the relative contribution of each arm remains an area of active investigation.
Clinical development proceeded through large phase 3 programmes in type 2 diabetes and in obesity or overweight with at least one weight-related comorbidity. Regulatory approvals followed in several jurisdictions for both indications. Weekly subcutaneous dosing reflects an elimination half-life of roughly five days. Open questions include the durability of metabolic effects after treatment stops, long-term cardiovascular and hepatic outcomes beyond completed trials, and whether the dual mechanism confers benefits independent of total receptor occupancy. Published literature continues to expand on these points. Substantial uncertainty remains about interindividual variability in response.
Development of tirzepatide took place under a research program that sought to test whether simultaneous engagement of two incretin receptors would produce greater metabolic effects than single-receptor agonism. Clinical trials were organized into the SURPASS series for type 2 diabetes and the SURMOUNT series for obesity and weight management. Regulatory clearance for type 2 diabetes came in 2022 in the United States, followed by approval for chronic weight management in 2023. The trial programs reported reductions in glycated hemoglobin and body weight relative to comparators, though long-term cardiovascular and durability data continue to accumulate.
The peptide backbone contains 39 amino acids and includes alpha-aminoisobutyric acid residues, which are not among the standard proteinogenic set. A C20 fatty diacid moiety is attached through a linker, allowing the compound to bind serum albumin and extend its circulation time. This albumin binding is the main reason the molecule supports once-weekly administration rather than more frequent dosing. The measured molecular mass is approximately 4,813 daltons, placing it firmly in the peptide rather than small-molecule class.

Long-term storage of lyophilised peptide powder is generally at minus twenty degrees Celsius or colder, with desiccant and protection from light. Short-term storage at two to eight degrees Celsius is common during active use. In solution, stability depends strongly on pH, concentration, and the presence of preservatives, and hydrolysis or aggregation can develop over weeks. Published stability data specific to this molecule are limited, so recommended conditions for research material are usually extrapolated from general peptide handling practice rather than from a dedicated study.
Bulk peptide material is normally characterised by reversed-phase high-performance liquid chromatography, which separates the target sequence from truncation products and other closely related impurities. Ultraviolet detection near 214 nanometres is common because the peptide backbone absorbs in that region. Mass spectrometry, usually electrospray ionisation coupled to a mass analyser, is used to confirm the molecular mass. Because the molecule carries a lipophilic side chain, gradient methods often need a relatively high organic modifier fraction to elute it within a practical retention window.
Like most synthetic peptides of this size, the material is commonly supplied as a lyophilised powder that appears white to off-white. It dissolves in aqueous buffers and in mixtures of water with a small proportion of organic solvent, though the fatty acid portion reduces solubility in pure water relative to short peptides. Hygroscopic behaviour is reported for many peptide powders, so weighing is usually performed quickly and under controlled humidity. Working solutions are typically prepared fresh and kept cold.
Verification of research-grade material involves checking purity, sequence and counter-ion content against a certificate of analysis. Reported purity figures usually reflect chromatographic area percentage and do not by themselves establish biological activity. Independent laboratories may repeat mass confirmation and peptide mapping to detect substitutions or truncations. Open questions concern how residual solvents, trace metals and subtle conformational variants affect measured behavior, and how consistently different suppliers define their specifications. Documentation of analytical methods matters as much as the headline purity number when results are compared across studies.
Routine characterization relies on reversed-phase high-performance liquid chromatography, often coupled to mass spectrometry, to confirm identity and estimate purity. Peptide mapping after enzymatic digestion verifies the amino acid sequence and locates appended groups such as the fatty acid chain. Size-exclusion chromatography detects aggregates and fragments, while ion-exchange chromatography resolves charge variants. Circular dichroism and nuclear magnetic resonance supply secondary and higher-order structural information in research settings. No single technique covers every attribute, so laboratories combine orthogonal methods and compare outcomes against a reference standard where one exists.
Development began in the 2010s, when researchers modified a GIP-based scaffold to add GLP-1 activity and then attached the fatty diacid to lengthen its half-life. Clinical evaluation proceeded through large phase 3 programmes in type 2 diabetes and in obesity, and regulators in the United States cleared the compound for type 2 diabetes in 2022 and for chronic weight management in 2023. Several cardiovascular and metabolic outcome studies are still reporting, so the picture of long-term benefit and risk is incomplete. Approvals in other regions followed on different timelines.
Tirzepatide is a synthetic peptide of 39 amino acids that carries a C20 fatty diacid side chain attached through a linker. Its molecular formula is C225H348N48O68, and its molecular weight is about 4813 daltons. The compound belongs to the incretin mimetic class and is administered by subcutaneous injection. The fatty acid chain promotes binding to serum albumin, which slows renal clearance and extends the circulation time of the molecule. It was identified during screening of sequences derived from glucose-dependent insulinotropic polypeptide.
Tirzepatide activates both the glucose-dependent insulinotropic polypeptide receptor and the glucagon-like peptide-1 receptor, making it a dual agonist rather than a selective agent. Engagement of the GLP-1 receptor is linked to glucose-dependent insulin release, slower gastric emptying, and reduced appetite signalling. The relative contribution of the GIP arm remains an active research question; proposed roles include improved insulin sensitivity and altered adipose tissue handling. Receptor occupancy studies suggest the molecule interacts with both targets at circulating concentrations achieved during therapy.
On 11 January, two Hezbollah-linked search-and-rescue personnel were killed and an unspecified number of civilians were wounded during an Israeli air raid on the town of Hanine. The IDF said that they killed four militants that infiltrated the Israeli border through Shebaa while an IDF unit was patrolling nearby on 14 January. Five IDF soldiers were wounded. A group calling itself the "Islamic Glory Brigades" later claimed responsibility for the attack and announced that 3 of their members were killed and 2 were able to escape. The same day, two Israeli civilians; a man and his 70-year-old mother, were killed by Hezbollah anti-tank missiles in Kfar Yuval. On 20 January, Brigadier General Sadegh Omidzadeh, an intelligence officer with the IRGC's Quds Force in Syria, along with four other IRGC officers, were reportedly killed in an Israeli airstrike in Damascus. According to the Syrian Observatory for Human Rights, an Israeli missile strike targeted a four-story building in the Mezzeh district. The attack resulted in the death of thirteen individuals, including five Iranians, and the complete destruction of the building where leaders aligned with Iran were having a meeting. The next day, in southern Lebanon, Sky News Arabic reported that Fadi Suleiman, a senior field commander of Hezbollah, survived an Israeli attack near a military checkpoint in Kafra. His bodyguard was killed in the same attack.
== Nature and significance == A quantity of radioactive waste typically consists of a number of radionuclides, which are unstable isotopes of elements that undergo decay and thereby emit ionizing radiation, which is harmful to humans and the environment. Different isotopes emit different types and levels of radiation, which last for different periods of time.
Later works of James Prescott Joule, Rudolf Clausius and in particular Ludwig Boltzmann firmly established the kinetic theory of gases and brought attention to both the theories of Bernoulli and Waterston. The debate between proponents of energetics and atomism led Boltzmann to write a book in 1898, which endured criticism until his suicide in 1906. Albert Einstein in 1905 showed how kinetic theory applies to the Brownian motion of a fluid-suspended particle, which was confirmed in 1908 by Jean Perrin.
== Television rights == The domestic broadcasting rights for the competition were held by the BBC, who had held them since the 2014–15 season, and ITV, who were showing matches for the second successive season. Both of these broadcasters will air the tournament until the 2024–25 season, and both were permitted to show the final live. In addition, FA Cup matches could also be streamed on ESPN+.

The price of real estate in Moscow continues to increase. The expected price averages $4,000 per square meter (11 sq ft) near the perimeter of the city or $6,500–8,000 per square meter in a prestigious district. The price sometimes exceeds $40,000 per square meter for a flat. Renting costs about $1,200 per month for a one-bedroom apartment and about $1,000 per month for a studio in the city center. A typical one-bedroom apartment in Moscow is about 30 square meters (320 square feet) in area; a typical two-bedroom apartment is 45 square meters (480 square feet); and a typical three-bedroom apartment is 70 square meters (750 square feet). Many residents cannot move out of their apartments, especially if a family lives in a two-room apartment originally granted by the state during the Soviet era. Some city residents have attempted to cope with the cost of living by renting out their apartments while staying in dachas (country houses) outside the city. In 2006, Mercer Human Resources Consulting named Moscow the world's most expensive city for expatriate employees, ahead of recurring winner Tokyo, because of the stable Russian ruble as well as increasing housing prices. Moscow also ranked first in the 2007 edition and 2008 edition of the survey. However, in 2009, Tokyo overtook Moscow, which came in third after Osaka. In 2014, according to Forbes magazine, Moscow was ranked the world's ninth most expensive city. This magazine ranked Moscow the second most expensive city the previous year.
=== Physiotherapy === For people with RA, physiotherapy may be used together with medical management. This may include cold and heat application, electronic stimulation, and hydrotherapy. Although medications improve symptoms of RA, muscle function is not regained when disease activity is controlled. Physiotherapy promotes physical activity. In RA, physical activity like exercise in the appropriate dosage (frequency, intensity, time, type, volume, progression) and physical activity promotion is effective in improving cardiovascular fitness, muscle strength, and maintaining a long-term active lifestyle. Additionally, exercise can be useful for pain management in this population, specifically, conditioning exercise programs that include aerobic, isometric, and isotonic exercises. Due to the debilitating effects of the disease, people with RA can gain skills back through exercise because it increases the energy capacity of the muscles. In the short term, resistance exercises, with or without range of motion exercises, improve self-reported hand functions. Physical activity promotion according to the public health recommendations should be an integral part of standard care for people with RA and other arthritic diseases. Additionally, the combination of physical activities and cryotherapy show its efficacy on the disease activity and pain relief.
=== Zubrin's planet mastery === In Entering Space: Creating a Spacefaring Civilization, Robert Zubrin suggests another form: his definition of a Type I civilization is described as one that has achieved full mastery of the resources of its planet (global), a Type II of its solar system (interplanetary), and a Type III would have unleashed the full potential of the galaxy (starfaring civilization). Metrics other than pure energy consumption have also been proposed. He ponders the possibility of a Type IV civilization, one that would dominate the universe, noting that there are limits to how minds can connect and interact on a galactic or intergalactic basis. As an example, he mentions that communication from the Galactic Center of the Milky Way galaxy to its edge would take about 50,000 years (since nothing can travel faster than light, according to our understanding of physics).
== Sequential and discrete models == Two kinds of models are usually used to represent protein samples: the sequential and the discrete (or non-sequential) models. The most elementary sequential model is to use the entire amino acid sequence, as expressed by:
Hypoxia occurs due to the decreased oxygen-binding capacity of methemoglobin, as well as the increased oxygen-binding affinity of other subunits in the same hemoglobin molecule, which prevents them from releasing oxygen at normal tissue oxygen levels.

Toxic Harmful; having to do with poison. Transcutaneous electric nerve stimulation (TENS) A treatment for painful neuropathy. Trauma Triglyceride Twenty-four hour urine The total amount of a person's urine for a 24-hour period. Diabetes mellitus type 1 It is a chronic condition in which the pancreas makes little or no insulin because the beta cells have been destroyed. About two-thirds of body cells require insulin to absorb glucose and in its absence, they will not be able to use the glucose (blood sugar) for energy. Type 1 diabetes usually comes on abruptly, although the damage to the beta cells may begin much earlier. Typical signs of Type 1 diabetes are a great thirst, hunger, a need to urinate often, and loss of weight. To treat the disease, the person must inject insulin and test blood glucose frequently. Type 1 diabetes usually occurs in children and adults who are under age 30. This type of diabetes used to be known as "insulin-dependent diabetes," "juvenile diabetes," "juvenile-onset diabetes" and "ketosis-prone diabetes." Diabetes mellitus type 2 The most common form of diabetes mellitus; about 90 to 95 percent of people who have diabetes in the developed world have Type 2 diabetes. Unlike Type 1 diabetes, in which the pancreas makes no insulin, people with Type 2 diabetes produce some insulin, sometimes even large amounts. However, either their bodies do not produce enough insulin or their body cells are resistant to insulin (see Insulin Resistance). People with Type 2 diabetes can often control their condition by losing weight through diet and exercise.
=== Low dispersion lenses === Thorium was also used in certain glass elements of Aero-Ektar lenses made by Kodak during World War II. Thus they are mildly radioactive. Two of the glass elements in the f/2.5 Aero-Ektar lenses are 11% and 13% thorium by weight. The thorium-containing glasses were used because they have a high refractive index with a low dispersion (variation of index with wavelength), a highly desirable property. Many surviving Aero-Ektar lenses have a tea colored tint, possibly due to radiation damage to the glass. These lenses were used for aerial reconnaissance because the radiation level is not high enough to fog film over a short period. This would indicate the radiation level is reasonably safe. However, when not in use, it would be prudent to store these lenses as far as possible from normally inhabited areas; allowing the inverse square relationship to attenuate the radiation.

=== Rebuild === In 2022, the Development Bank of Southern Africa (DBSA) was appointed to lead a rebuild of the parliament buildings. An initial estimate of R2.2 billion for the rebuild in 2024 ballooned to R4.6 billion in 2025. The rebuild includes the 1983 National Assembly building, the Old Assembly Building, and the 90 Plein Street offices. In August 2026, members of the Joint Standing Committee on the Financial Management of Parliament visited Parliament's construction site to assess the rebuild's progress. During a committee meeting, it was noted that the project was still within budget, with R2.43 billion of the total R4.68 billion having been spent so far. The rebuild was on track to finish by its target date of June 2027, before that year's SONA. According to the project's implementing agent, the Development Bank of Southern Africa (DBSA), construction of the new National Assembly building was 76% complete at the time.
The clinical course is determined by an immunological interplay between two types of T cells (Th1 and Th2 responses). SCD is often considered a subset of SNAS, but with only skin manifestations. SNAS presents with an array of symptoms ranging from respiratory to generalized skin rash to gastrointestinal symptoms. The gastrointestinal symptoms may mimic those of irritable bowel syndrome. A meta review evaluating SNAS found that 1% of patients sensitized to nickel reacted to the nickel content of a 'normal' diet, and with increasing doses of nickel more individuals reacted. SNAS is a multilayered immunological response demonstrating variance between individuals and doses of nickel exposure.

It is a dual GIP and GLP-1 receptor agonist, frequently grouped with incretin-based peptide therapeutics. It is a peptide rather than a small molecule and is given by subcutaneous injection.
Selective agents engage only the GLP-1 receptor, whereas tirzepatide activates GIP and GLP-1 receptors simultaneously. This difference in receptor coverage is the principal pharmacological distinction emphasised in comparative reviews.
Downstream signalling is partly characterised, but the quantitative contribution of GIP versus GLP-1 receptor activation to metabolic outcomes is not settled. Review articles commonly flag this as an unresolved question rather than a settled finding.
It binds and activates both the GIP and GLP-1 receptors, making it a dual incretin receptor agonist. Single-receptor GLP-1 agonists act on one target only. The dual profile is the defining pharmacological feature of the molecule.
