Tirzepatide is a synthetic peptide built from 39 amino acid residues. Its sequence is related to human glucose-dependent insulinotropic polypeptide, with modifications that include a C-terminal extension and a C20 fatty diacid joined through a linker. Those changes raise the molecule's affinity for serum albumin, which slows renal filtration and lengthens the time it stays in circulation. The free base has an average molecular mass near 4813.5 daltons. The compound is made by solid-phase peptide synthesis followed by chromatographic purification.
At the receptor level, tirzepatide activates both the glucose-dependent insulinotropic polypeptide receptor and the glucagon-like peptide-1 receptor. Both belong to the class B family of G protein-coupled receptors and signal largely through cyclic AMP accumulation. The compound binds the two receptors with differing affinity, and the pattern of signaling at each site is described in the literature as biased rather than simply proportional to occupancy. Tissues carrying these receptors include pancreatic islets, adipose tissue, the central nervous system, and the gastrointestinal tract. The relative weight of each receptor population in producing metabolic effects continues to be studied.
Identity and purity are usually established with reversed-phase high-performance liquid chromatography for the main peak and with mass spectrometry for the observed molecular mass. Peptide mapping after enzymatic digestion confirms the primary sequence, while amino acid analysis provides a quantitative composition check. Size-exclusion chromatography and ion-exchange chromatography are used to look for aggregates and charge variants. Water content, residual solvents, and counter-ion content are measured separately, since a lyophilised powder is often reported on an as-is basis unless a correction is applied.
Research-grade material circulates through suppliers that differ widely in documentation and testing practice, so a certificate of analysis is a starting point rather than proof of quality. Independent verification typically repeats chromatographic purity and mass confirmation on the received lot, and compares results against a retained reference standard. Regulatory status varies by jurisdiction, and a substance cleared as a medicine is not interchangeable with a research chemical of the same name. Open questions include how closely non-pharmaceutical lots match approved material in impurity profile and in aggregate content.

Structure-activity work shows that fatty acid length, linker chemistry and the position of acylation all influence albumin affinity and receptor potency. Plasma protein binding exceeds 99 percent, which restricts distribution and slows renal clearance. Degradation proceeds largely through general proteolysis and fatty acid oxidation rather than cytochrome P450 metabolism, so exposure to common oxidative drug interactions is limited. Whether these clearance routes vary meaningfully between individuals is not fully established.
The molecule is a synthetic 39-amino-acid peptide whose backbone derives from the sequence of human glucose-dependent insulinotropic polypeptide, with several substitutions that raise metabolic stability and shift receptor preference. A C20 fatty diacid is attached through a short linker to a lysine side chain, a modification that increases binding to serum albumin. The reported monoisotopic mass is approximately 4813 Da. Near neutral pH the peptide carries a net negative charge, and the lipid tail makes the molecule markedly more hydrophobic than the unmodified parent sequence.
A through-gut (one with both mouth and anus) is thought to have evolved within the nephrozoan clade of Bilateria, after their ancestral ventral orifice (single, as in cnidarians and acoels; re-evolved in nephrozoans like flatworms) stretched antero-posteriorly, before the middle part of the stretch would get narrower and closed fully, leaving an anterior orifice (mouth) and a posterior orifice (anus plus genital opening). A stretched gut without the middle part closed is present in another branch of bilaterians, the extinct proarticulates. This and the amphistomic development (when both mouth and anus develop from the gut stretch in the embryo) present in some nephrozoans (e.g. roundworms) are considered to support this hypothesis.
== Medically significant species == Mortality (often determined by measured toxicity on mice) is a commonly used indicator of the danger of any given venomous snake, but important too are efficiency of venom delivery, venom yield, and behavior upon encountering humans. Given these additional, practical concerns, many snake experts have cited the black mamba and the coastal taipan as the world's most dangerous snakes – even though their venoms, drop for drop, are not the most lethal. Both species are elapids, and in several aspects of morphology, ecology and behavior, the coastal taipan is strongly convergent with the black mamba.

Israel is one of only a few nations that conscript women or deploy them in combat roles. In practice, women can avoid conscription through a religious exemption and over a third of Israeli women do so. As of 2010, 88% of all roles in the IDF are open to female candidates, and women were found in 69% of all IDF positions. According to the IDF, 535 female Israeli soldiers were killed in combat operations in the period 1962–2016, and dozens before then. The IDF says that fewer than 4 percent of women are in combat positions. Rather, they are concentrated in "combat-support" positions which command a lower compensation and status than combat positions.
Expansion of the bone marrow in the developing child leads to a distinctive facial shape often referred to as "Chipmunk facies". Other skeletal changes include osteoporosis, growth retardation, and malformation of the spine. People with thalassemia can get too much iron in their bodies, either from the disease itself as RBCs are destroyed, or as a consequence of frequent blood transfusions. Excess iron is not excreted, but forms toxic non-transferrin-bound iron. This can lead to organ damage, potentially affecting the heart, liver, endocrine system, bones and spleen. Symptoms include an irregular heartbeat, cardiomyopathy, cirrhosis of the liver, hypothyroidism, delayed puberty and fertility problems, brittle and deformed bones, and an enlarged spleen. The spleen is the organ which removes damaged red blood cells from circulation; in thalassemia patients it is abnormally active, causing it to enlarge and possibly become hyperactive, a condition called hypersplenism. The immune system can become compromised in a number of ways; anemia, iron overload, and hypersplenism may affect the immune response and increase the risk of severe infection.
This contradicts information published earlier in a Wired article, which states that the hyperthymesiac Jill Price had been brain scanned and her "hippocampus and prefrontal cortex were reportedly normal", suggesting that these regions of the brain do not need to be different for hyperthymesia to occur. Significant debate also exists over the limits of memory capacity. Some are of the view that the brain contains so many potential synaptic connections that, in theory at least, no practical limit exists to the number of long-term memories that the brain can store. In 1961, Wilder Penfield reported that specific stimulation of the temporal lobes resulted in vivid recollection of memories. He concluded that our brains were making "continuous, effortless, video-like recordings" of our experiences, but that these records are not consciously accessible to us. However, a study published in the Proceedings of the National Academy of Sciences suggested that hyperthymesiacs may reconstruct memories from traces and incorporate post-event information and associations—a finding at odds with Penfield's video-like recording analogy.
Education and health are provided mainly by member states, but shaped by common minimum standards in EU law. In the case of education, the European Social Charter, like the Universal Declaration, the International Bill of Human Rights, say that "everyone" has the right to education, and that primary, secondary and higher education should be made "free", for instance "by reducing or abolishing any fees or charges" and "granting financial assistance". While the history of education was confined to a wealth elite, today most member states have tuition free university. There are no common rules for university finance or governance, although there is a right to free movement and universities have voluntarily harmonised standards. In 1987, the Erasmus Programme was created to fund students to study in other countries, and with a budget of €30 billion from 2021 to 2027. From 1999, the Bologna Declaration and Process led to the creation of the European Higher Education Area where member state universities adopted a common degree structure (bachelor, master, and doctoral degree) with a goal to have similar expectations in learning outcomes. Member states may not impose different fees on students from other member states or limit their numbers, and this appears to have worked even without a system for countries to reimburse one another if costs differ widely. However, if member states have grants or student loans, R (Bidar) v London Borough of Ealing held there may be a minimum residency requirement, such as three years.
DM is paraneoplastic in up to 40% of cases, most commonly in association with underlying malignancy in patients with anti–TIF1γ autoantibodies. A majority of patients with paraneoplastic dermatomyositis harbor somatic mutations or other genetic alterations in tumor genes encoding the target autoantigens of their corresponding autoantibodies, suggesting that, in this context, autoantibody production arises secondary to an anti-tumor immune response that fails to achieve effective tumor control. The most commonly associated cancers are ovarian cancer, breast cancer, and lung cancer overall, but the most frequent associations can vary depending on patient race or ethnicity. Inherited genetic factors confer a predisposition to developing these diseases, and HLA subtypes HLA-DR3, HLA-DR52, and HLA-DR6 seem to create a disposition to DM.

Trials are investigating whether botulinum toxin, when used to make a person appear to frown less, stops negative feedback from the face and affects depression. Psilocybin may have a beneficial role in the treatment of depression. A 2019 meta-analysis found that hypnotherapy may be an effective way of alleviating the symptoms of depression. No model of depression in animals that fully explains the mechanism of depression has been found as of 2019.
Controversy exists as to whether some of these treatments should be acceptable, but one can be more or less forgiven universally: This is the light waxing or oiling applied to most gem turquoise to improve its colour and lustre; if the material is of high quality to begin with, very little of the wax or oil is absorbed and the turquoise therefore does not rely on this impermanent treatment for its beauty. All other factors being equal, untreated turquoise will always command a higher price. Bonded and reconstituted material is worth considerably less. Being a phosphate mineral, turquoise is inherently fragile and sensitive to solvents; perfume and other cosmetics will attack the finish and may alter the colour of turquoise gems, as will skin oils, as will most commercial jewellery cleaning fluids. Prolonged exposure to direct sunlight may also discolour or dehydrate turquoise. Care should therefore be taken when wearing such jewels: cosmetics, including sunscreen and hair spray, should be applied before putting on turquoise jewellery, and they should not be worn to a beach or other sun-bathed environment. After use, turquoise should be gently cleaned with a soft cloth to avoid a buildup of residue, and should be stored in its own container to avoid scratching by harder gems. Turquoise can also be adversely affected if stored in an airtight container.
A downside of NFT is that it has very little buffering against interruptions in the flow (e.g., power outages). But, overall, it is probably one of the more productive techniques. The same design characteristics apply to all conventional NFT systems. While slopes along channels of 1:100 have been recommended, in practice it is difficult to build a base for channels that is sufficiently true to enable nutrient films to flow without ponding in locally depressed areas. As a consequence, it is recommended that slopes of 1:30 to 1:40 are used. This allows for minor irregularities in the surface, but, even with these slopes, ponding and water logging may occur. The slope may be provided by the floor, benches or racks may hold the channels and provide the required slope. Both methods are used and depend on local requirements, often determined by the site and crop requirements. For nutrient film technique (NFT) systems, recommended flow rates are commonly around 1 L/min per gully to provide sufficient nutrient replenishment to the root film. At planting, rates may be half this and the upper limit of 2 L/min appears about the maximum. Flow rates beyond these extremes are often associated with nutritional problems. Depressed growth rates of many crops have been observed when channels exceed 12 meters in length. On rapidly growing crops, tests have indicated that, while oxygen levels remain adequate, nitrogen may be depleted over the length of the gully. As a consequence, channel length should not exceed 10–15 meters.
Only after a fierce debate, considered by many as one of the most memorable sessions of parliament, the Bundestag concluded on 20 June 1991, with quite a slim majority, that both government and parliament should move to Berlin from Bonn.

It is a synthetic peptide and a dual agonist of two incretin receptors. It is not a small molecule, and it is not structurally related to the older single-receptor peptide agonists.
The C20 fatty diacid promotes tight binding to serum albumin. That binding reduces renal clearance and extends circulation time compared with an unmodified peptide of similar length.
It is not fully established. Studies indicate that both receptors contribute to the observed effects, but the exact split between the two signaling pathways in humans remains an open question.
It is normally kept frozen, desiccated, and away from light, with brief warming to room temperature before opening to limit condensation. Repeated freeze-thaw cycles are avoided because they stress the peptide. Once in solution, the material is held cold and used promptly.
