Tirzepatide is a synthetic peptide of 39 amino acids that carries a C20 fatty diacid side chain attached through a linker. Its molecular formula is C225H348N48O68, and its molecular weight is about 4813 daltons. The compound belongs to the incretin mimetic class and is administered by subcutaneous injection. The fatty acid chain promotes binding to serum albumin, which slows renal clearance and extends the circulation time of the molecule. It was identified during screening of sequences derived from glucose-dependent insulinotropic polypeptide.
Tirzepatide activates both the glucose-dependent insulinotropic polypeptide receptor and the glucagon-like peptide-1 receptor, making it a dual agonist rather than a selective agent. Engagement of the GLP-1 receptor is linked to glucose-dependent insulin release, slower gastric emptying, and reduced appetite signalling. The relative contribution of the GIP arm remains an active research question; proposed roles include improved insulin sensitivity and altered adipose tissue handling. Receptor occupancy studies suggest the molecule interacts with both targets at circulating concentrations achieved during therapy.
Like most synthetic peptides of this size, the material is commonly supplied as a lyophilised powder that appears white to off-white. It dissolves in aqueous buffers and in mixtures of water with a small proportion of organic solvent, though the fatty acid portion reduces solubility in pure water relative to short peptides. Hygroscopic behaviour is reported for many peptide powders, so weighing is usually performed quickly and under controlled humidity. Working solutions are typically prepared fresh and kept cold.
Long-term storage of lyophilised peptide powder is generally at minus twenty degrees Celsius or colder, with desiccant and protection from light. Short-term storage at two to eight degrees Celsius is common during active use. In solution, stability depends strongly on pH, concentration, and the presence of preservatives, and hydrolysis or aggregation can develop over weeks. Published stability data specific to this molecule are limited, so recommended conditions for research material are usually extrapolated from general peptide handling practice rather than from a dedicated study.

Tirzepatide is a synthetic linear peptide of 39 amino acids that acts as a dual agonist at the glucose-dependent insulinotropic polypeptide (GIP) and glucagon-like peptide-1 (GLP-1) receptors. Its sequence derives from native GIP but incorporates non-natural residues and a C20 fatty diacid moiety linked to a lysine side chain. The lipophilic chain promotes albumin binding, which slows renal clearance and extends circulation time. The unmodified peptide has a molecular formula of C225H348N48O68 and a molecular mass near 4,813 daltons.
Receptor activation by tirzepatide raises intracellular cyclic AMP through Gs-coupled signalling at both targets. At the GLP-1 receptor the downstream effect includes glucose-dependent insulin release, suppressed glucagon secretion, delayed gastric emptying, and reduced appetite signalling in the hypothalamus. GIP receptor engagement adds insulinotropic activity and appears to influence lipid handling in adipose tissue. Because both receptors are stimulated at the same time, the pharmacological profile differs from that of selective GLP-1 receptor agonists, and the relative contribution of each arm remains an area of active investigation.
Clinical development proceeded through large phase 3 programmes in type 2 diabetes and in obesity or overweight with at least one weight-related comorbidity. Regulatory approvals followed in several jurisdictions for both indications. Weekly subcutaneous dosing reflects an elimination half-life of roughly five days. Open questions include the durability of metabolic effects after treatment stops, long-term cardiovascular and hepatic outcomes beyond completed trials, and whether the dual mechanism confers benefits independent of total receptor occupancy. Published literature continues to expand on these points. Substantial uncertainty remains about interindividual variability in response.
Most pesticides protect crops against insects and plants, but in some cases antimicrobial pesticides are used to protect against various microorganisms such as bacteria, viruses, fungi, algae, and protozoa. The overuse of many pesticides in an effort to have a higher yield of crops has resulted in many of these microbes evolving a tolerance against these antimicrobial agents. Currently there are over 4000 antimicrobial pesticides registered with the US Environmental Protection Agency (EPA) and sold to market, showing the widespread use of these agents. It is estimated that for every single meal a person consumes, 0.3 g of pesticides is used, as 90% of all pesticide use is in agriculture. A majority of these products are used to help defend against the spread of infectious diseases, and hopefully protect public health. But out of the large amount of pesticides used, it is also estimated that less than 0.1% of those antimicrobial agents, actually reach their targets. That leaves over 99% of all pesticides used available to contaminate other resources. In soil, air, and water these antimicrobial agents are able to spread, coming in contact with more microorganisms and leading to these microbes evolving mechanisms to tolerate and further resist pesticides. The use of antifungal azole pesticides that drive environmental azole resistance have been linked to azole resistance cases in the clinical setting. The same issues confront the novel antifungal classes (e.g. orotomides) which are again being used in both the clinic and agriculture.
=== In medicine === Many hospitals have pneumatic tube systems which send samples to laboratories. Blood preservations are transported, where weight and transport duration matter as well as preventing haemolysis caused by centrifugal and accelerating forces. Pneumatic tube systems are also used in hospitals to transport X-rays, patient documents, general documents, drugs and test results. 6-inch (152 mm) pneumatic tube systems have been shown to handle heavy liter-capacity IV bags with significantly fewer jams compared to the 4-inch (102 mm) systems.
=== Classification === Historical classification systems were based on clinical factors. Until the early 1970s, the idea of a single amyloid substance predominated. Various descriptive classification systems were proposed based on the organ distribution of amyloid deposits and clinical findings. Most classification systems included primary (i.e., idiopathic) amyloidosis, in which no associated clinical condition was identified, and secondary amyloidosis (i.e., secondary to chronic inflammatory conditions). Some classification systems included myeloma-associated, familial, and localized amyloidosis. The modern era of amyloidosis classification began in the late 1960s with the development of methods to make amyloid fibrils soluble. These methods permitted scientists to study the chemical properties of amyloids. Descriptive terms such as primary amyloidosis, secondary amyloidosis, and others (e.g., senile amyloidosis), which are not based on cause, provide little useful information and are no longer recommended. The modern classification of amyloid disease tends to use an abbreviation of the protein that makes the majority of deposits, prefixed with the letter A. For example, amyloidosis caused by transthyretin is termed "ATTR". Deposition patterns vary between people but are almost always composed of just one amyloidogenic protein. Deposition can be systemic (affecting many different organ systems) or organ-specific. Many amyloidoses are inherited, due to mutations in the precursor protein.

In prehistoric times, they have been used for making bone tools. They have further been used in bone carving, already important in prehistoric art, and also in modern time as crafting materials for buttons, beads, handles, bobbins, calculation aids, head nuts, dice, poker chips, pick-up sticks, arrows, scrimshaw, and ornaments. Bone glue can be made by prolonged boiling of ground or cracked bones, followed by filtering and evaporation to thicken the resulting fluid. Once historically important, bone glue and other animal glues today have only a few specialized uses, such as in antiques restoration. Essentially the same process, with further refinement, thickening and drying, is used to make gelatin. Broth is made by simmering several ingredients for a long time, traditionally including bones. Bone char, a porous, black, granular material primarily used for filtration and also as a black pigment, is produced by charring mammal bones. Oracle bone script was a writing system used in ancient China based on inscriptions in bones. Its name originates from oracle bones, which were mainly ox clavicle. The Ancient Chinese (mainly in the Shang dynasty), would write their questions on the oracle bone, and burn the bone, and where the bone cracked would be the answer for the questions. The wishbones of fowl have been used for divination, and are still customarily used in a tradition to determine which one of two people pulling on either prong of the bone may make a wish.

Largely destroyed by the Allies during the Second World War, the city was rebuilt according to the plans of the architect Auguste Perret between 1945 and 1964. Only the City Hall and the Church of Saint Joseph (107-metre high (351 ft)) were personally designed by Auguste Perret. In commending the reconstruction work UNESCO listed the city of Le Havre on 15 July 2005 as a World Heritage Site. This area of 133 hectares (330 acres) is one of the few inscribed contemporary sites in Europe. The architecture of the area is characterized by the use of precast concrete using a system of a modular frame of 6.24 m (20.5 ft) and straight lines. Another notable architectural work of the central city is that of the House of Culture built in 1982 by the Brazilian architect Oscar Niemeyer and nicknamed "the Volcano" because of the shape of the building. From 2012, this place was refurbished both inside and outside with fairly significant changes approved by the architect including greater openness to the outside of the plaza. The Notre Dame and Perrey neighbourhoods are mainly residential. Les Halles is one of the commercial hubs of the city. The Saint Francis neighborhood was also rebuilt beginning in 1950 but in a radically different architectural style: the buildings are brick and have pitched slate roofs. This is the restaurant district and the fish market.
== Advantages == The B1H system has significant advantages over other methods that investigate protein–DNA interactions. Microarray-based readout of chromatin immunoprecipitation (ChIP-chip) for high-throughput binding-site determination relies on specific antibodies which may not always be available. Methods that rely on protein-binding microarrays also require additional protein purification steps that are not required in the B1H system. Furthermore, these microarray-based techniques are often prohibitive in terms of requiring special facilities and expertise to analyze the resulting data. SELEX, another system commonly used to identify the target nucleic acids for DNA-binding proteins, requires multiple rounds of selection. In contrast, the bacterial one-hybrid system requires just one round of in vitro selection and also offers a low-tech alternative to microarray-based technologies. Antibodies are not required for studying the interactions of DNA-binding proteins in the B1H system. A further advantage is that the B1H system works not only for monomeric proteins but also for proteins that bind DNA as complexes. The B1H system should be considered a specialized technique for studying DNA-protein interactions whereas the two-hybrid variations (B2H and Y2H) can assess both protein–protein and protein–DNA interactions. These two-hybrid systems are multi-purpose but are limited in terms of assaying only a single “prey” library.
=== Mitigation measures === Several strategies have been proposed as a way to protect people at the greatest risk of occupational exposure to PFAS, including exposure monitoring, regular blood testing, and the use of PFAS-free alternatives such as fluorine-free firefighting foam and plant-based ski wax.
=== 4 June === One person was killed in a Russian attack in Kherson Oblast. Another was killed by shelling in Sumy Oblast. The SBU arrested a man in Zaporizhzhia Oblast on suspicion of plotting an attack against a military enlistment office on behalf of Russia. Russia "vehemently denied" any involvement in an event in which five coffins draped in French flags were placed under the Eiffel Tower in Paris with the inscription "French soldiers of Ukraine". French media claimed that it was a Kremlin attempt to weaken French support for Ukraine. Ukraine announced plans to establish a "decentralised energy system" using "mini-power plants" to reduce vulnerability to Russian attacks after the country's generating capacity was found to have been reduced to below 20 gigawatts from its previous capacity of 55 gigawatts.

It is a synthetic 39-amino-acid peptide that acts on two incretin receptors, the GIP receptor and the GLP-1 receptor. It is given by subcutaneous injection and has a circulating half-life of roughly five days. It is not a small molecule and is not absorbed usefully from the gut in conventional oral form.
Selective agents act on one receptor, while tirzepatide engages both GIP and GLP-1 receptors. The added GIP activity may contribute effects on insulin sensitivity and on fat metabolism. Whether the dual action produces meaningful clinical advantages beyond differences in potency is still being examined.
The downstream consequences of GIP receptor activation are not fully characterised in humans. It is also unclear how much each receptor contributes to appetite reduction and to shifts in body composition. Published work describes associations and proposed pathways rather than settled causal chains.
It separates molecules by hydrophobicity, which is effective for distinguishing an intact peptide from truncated or chemically modified forms. A C18 column with an acidic water-organic mobile phase is a standard configuration.
